Transcription factor translated in axons

چکیده
F ilters in the mammalian kidney must be unclogged by a transport receptor, say Shreeram Akilesh, Andrey Shaw (Washington University, St. Louis, MO), and colleagues. The receptor moves obstructing proteins into the urine, where they can later be returned to the bloodstream. The kidney fi lters the blood through a bed of capillaries known as the glomerulus. Large and highly charged proteins are mostly kept out of the urine fi ltrate fi rst by a basement membrane and then by fi nger-like projections of epithelial cells called podocytes. Shaw’s group wondered why the gaps between podocyte projections do not get clogged by the proteins they prevent from passing. The two dominant large proteins in the blood— those mostly likely to cause a clog—are albumin and IgG. Using microarray analyses, the team found that a receptor for these proteins, called FcRn, was expressed in podocytes. FcRn was fi rst identifi ed for its role in bringing IgG antibodies from breast milk across the surface of an infant’s gastrointestinal tract. The new fi ndings suggest it similarly transports IgG and albumin across the podocyte surface into the urinary fl uid. In mice lacking FcRn, IgG clearance was slowed, and the antibody accumulated in the kidney fi lter. Clogged kidneys meant greater susceptibility to agents that would otherwise be quickly cleared; a normally harmless dose of a kidney-targeting antibody was toxic to mice whose kidneys were overloaded with IgG. An overloaded fi lter might also explain why kidney failure is often the downfall of patients with lupus. “The autoantibodies in lupus patients aren’t so uncommon,” says Shaw. “But with clogging from all the excess antibodies, the kidneys probably cannot get cleared of them in time.” Because IgG and albumin are not normally found in urinary waste, the authors imagine that they are reabsorbed into the bloodstream in kidney tubules, where sugars and amino acids are also reclaimed. The expression of FcRn in tubules supports this idea. To prove their theory, the authors hope to put FcRn back into podocytes but not tubules of the knockout mice, to see whether IgG is sent out with the urine. Reference: Akilesh, S., et al. 2008. Proc. Organization and translation of mRNA in sympathetic axons.
Many axons carry out the synthesis of macromolecules independent of their cell bodies but the nature, organization and magnitude of axonal protein synthesis remain unclear. We have examined these features in axons of chick sympathetic neurons in cell culture. In situ hybridization showed that poly(A) mRNA is abundant and non-uniformly distributed in nearly all axons. The specific transcripts fo...
متن کاملTarget-derived neurotrophins coordinate transcription and transport of bclw to prevent axonal degeneration.
Establishment of neuronal circuitry depends on both formation and refinement of neural connections. During this process, target-derived neurotrophins regulate both transcription and translation to enable selective axon survival or elimination. However, it is not known whether retrograde signaling pathways that control transcription are coordinated with neurotrophin-regulated actions that transp...
متن کاملFGF as a Target-Derived Chemoattractant for Developing Motor Axons Genetically Programmed by the LIM Code
LIM transcription factors confer developing axons with specific navigational properties, but the downstream guidance receptors and ligands are not well defined. The dermomyotome, a transient structure from which axial muscles arise, is the source of a secreted long-range chemoattractant specific for medial-class spinal motor neuron axons (MMCm axons). We show that fibroblast growth factors (FGF...
متن کاملDevelopment/Plasticity/Repair R7 Photoreceptor Axon Growth Is Temporally Controlled by the Transcription Factor Ttk69, Which Inhibits Growth in Part by Promoting Transforming Growth Factor- /Activin Signaling
Work on axon growth has classically focused on understanding how extrinsic cues control growth cone dynamics independent of the cell body. However, more recently, neuron-intrinsic transcription factors have been shown to influence both normal and regenerative axon growth, suggesting that understanding their mechanism of action is of clinical importance. We are studying axon targeting in the Dro...
متن کاملP 129: The Role of Overexpression Transcription Factor BRN 4 in Multiple Sclerosis
Adult neurogenesis is a process of producing nerve cells from their progenitor that occurs in some areas in the brain such as the hypothalamus. Low activity in this area plays a role in neural degeneration and diseases such as multiple sclerosis, epilepsy and depression. MS is a neurodegenerative disease with a permanent disability that the main reason for it is axonal degeneration and neuronal...
متن کاملImproving Rice (Oryza sativa L.) Drought Tolerance by Suppressing a NF-YA Transcription Factor
The response to drought stress is a complicated process involving stress sensing, intracellular signaltransduction, and the execution of a cellular response. Transcription factors play important roles in the signaling pathways including abiotic stress. In the present study a rice NF-YA transcription factor gene was partially characterized following dehydration. Disrupting the gene via a T...
